The weapon wheel and vehicle storage

You cannot carry an armoury any more. A sidearm is what fits on you. Anything bigger travels in the car.

The bottomless weapon wheel is gone. Footage from the Extended Look shows larger weapons living in your vehicle and being pulled out when you need them, with what you can carry on your person reduced to something closer to plausible.

The car is now equipment

This is the change with the widest blast radius. In Grand Theft Auto V your car was transport and cover. Here it is also your armoury, which means every decision about the vehicle becomes a decision about your loadout. Parking badly costs you. Losing the car during a chase costs you more than a walk home. Choosing to arrive on a bike is choosing what you can bring.

It also gives the evidence system real teeth. Your weapon is a tracked item and your vehicle is a tracked item, and now the two travel together. Ditching a car that police have a description of means ditching what is inside it, or spending time at the boot in the open while they close.

Series context

Carry limits are not new to Rockstar. Red Dead Redemption 2 restricted you to what was on your horse and on your back, and the horse becoming a genuine piece of equipment was one of the things people remembered about it. This is the same idea rebuilt around a car, in a game where cars are stolen and abandoned constantly rather than raised and kept.

What is not known

How many weapons a vehicle holds, whether the storage is per vehicle or follows you, and what happens to weapons in a car the police impound have not been shown. Nor has Rockstar confirmed how large the on-person limit is.
